Sci. Adv.: Eyedrop-based macromolecular ophthalmic drug delivery for ocular fundus disease treatment

Fundus diseases have been widely concerned due to their increasing incidence, sensory impairment, limited treatment and high postoperative risk.Nowadays, therapeutic antibodies have been widely used in intravitreal injection to treat fundus diseases and have achieved certain clinical efficacy.However, intravascular injection can cause discomfort to patients, and there may be risks of intraocular infection, intraocular bleeding, and retinal detachment.Therefore, there is an urgent need to develop an efficient local ophthalmic drug delivery system for non-invasive delivery of therapeutic macromolecule drugs to fundus patients. However, the complex ocular barrier structure often limits the research and development of most preparations.

 

In response to this problem, Prof. Liu Zhuang and Prof. Chen Qian from the Institute of Functional Nano and Soft Materials of Soochow University, Prof. Gu Ping from the ophthalmic Center of the Ninth People's Hospital of Shanghai Jiao Tong University and Prof. Zhang Xiao-feng from the Ophthalmic Center of Dushu Lake Hospital of Soochow University published a paper entitled Eyedrop-based macromolecular ophthalmic drug delivery for ocular fundus disease treatment .The results report a novel eye barrier penetration vector based on fluorocarbon chain modified chitosan (FCS) for the delivery of large molecular ophthalmic drugs in eye drop formulations. This strategy can effectively cross complex eye structures to the fundus of mice and rabbits for the treatment of fundus tumors or choroidal neovascularization (CNV).The results have been published in Science Advances (DOI: 10.1126/sciadv.abq3104).Shen Jingjing, PhD student of Professor Chen Qian's research group, is one of the co-first authors of this paper.
